Background of alpha Adaptin

The AP2A1 and AP2A2 represent the AP2 alpha proteins that are found in the AP2 complex in clathrin-coated vesicles. AP2A is also known as Alpha-adaptin A, Adaptor protein complex AP-2 alpha-1 subunit, Clathrin assembly protein complex 2 alpha-A large chain, 100 kDa coated vesicle protein A and Plasma membrane adaptor HA2/AP2 adaptin alpha A subunit. The AP2 complex is a heterotetramer consisting of two large adaptins (alpha or beta), a medium adaptin (mu), and a small adaptin (sigma). The complex is part of the protein coat on the cytoplasmic face of coated vesicles, which link clathrin to receptors in vesicles.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-AP2A antigen affinity purified polyclonal antibody (Catalog # A30987) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a Rabbit anti-Goat IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1023) with Tanon 5200 system. A specific band was detected for AP2A. test
